METHOD FOR EXPRESSION CLONING OF GENE CODING FOR LIGAND BINDING PROTEIN

PROBLEM TO BE SOLVED: To provide an efficient method for cloning a DNA coding for a ligand- binding protein. SOLUTION: A DNA inserted into an expression vector is transduced into a host. The host is cultured on a plate and the expressed protein is transferred from the plate to a film by bringing a film into contact with the surface of the plate. The film is peeled off from the plate and a labeled ligand is added to the film to effect the binding of the ligand to the protein. The label bonded to the protein is detected and a DNA is separated from the clone on the plate corresponding to the positive position of the film.
