FIELD: food and medicinal industry. SUBSTANCE: method involves raw milling, preliminary extraction of total lipids, repeated extraction of solid deposit, filtration of the combined extracts and evaporation, multiple cholesterol extraction from fat and cholesterol crystallization. Preliminary extraction is carried out with a mixture of chloroform and ethanol at ratio raw : chloroform : ethanol = 1:2:2. Obtained solid deposit is extracted repeatedly with ethanol at ratio ethanol : deposit = 5:1. Multiple extraction of cholesterol from fat is carried out with a mixture ethanol : acetone at ratio (2:0.5)-(3:1) at ratio fat : mixture = (1:3)-(1:5). Combined extracts were filtered, evaporated and dissolved in hot ethanol at below 75 C at ratio deposit : ethanol = 1:10, cooled for 3 h at (-5 C)-(-10 C) and filtered. Solid deposit is washed with a mixture alcohol : acetone at ratio = 3:1 at ratio deposit : mixture = 1:5. Cholesterol is crystallized at -5 C (not above) for 3-5 h. EFFECT: higher efficiency. |
