PROBLEM TO BE SOLVED: To obtain the subject vaccine comprising an immunogenic complex of iscom structure obtained by applying a specific treatment to an amphiphatic antigenic protein and exhibiting a stronger immunogenic power than that of a composition wherein the antigenic protein is contained in the form of aggregation. SOLUTION: This vaccine is obtained by bringing an amphiphatic antigenic protein or peptide in a dissolved or solubilized form into contact with a solution containing (A) a detergent (preferably octyl glycoside), (B) a sterol (preferably cholesterol), (C) a phospholipid (preferably phosphatidylethanolamine) and (D) a glycoside comprising hydrophobic and hydrophilic regions in at least the critical micelle-forming concentration (preferably Quil A extracted from the bark of Quillaja saponarta), removing the component A from the mixture preferably by a gel filtration method, etc., purifying the produced immunogenic complex, and finally adding the immunogenic complex to a medium generally comprising a buffering agent, a physiological salt solution, etc.
