<p>PURPOSE: To provide a new monoclonal antibody capable of inhibiting blood coagulation and recognizing neutral glycolipids, having high specificity to malignant tumor, exhibiting high sensitivity even to early malignant tumor and useful e.g. as an agent for detecting malignant tumor by using blood, urine, tissue, etc., as a specimen. CONSTITUTION: A female BALB/c mouse of 8 weeks old is immunized by injecting LK-52 cell originated from human squamous cell carcinoma into the peritoneal cavity of the mouse 4 times with 2 weeks interval. Spleen cell is collected from the mouse 3 days after the final injection and fused with a mouse myeloma cell using polyethylene glycol. The fused cell is selectively cultured on a HAT medium to obtain a hybridoma. The product is screened by using a neutral glycolipid extracted from LK-52 cell originated from human squamous cell carcinoma to select a strain producing an antibody having specificity to neutral glycolipid. The strain is cloned by limited dilution method, etc., and the obtained monoclonized hybridoma is cultured to obtain the objective new monoclonal antibody cable of inhibiting the coagulation of blood and recognizing neutral glycolipids.</p> |
