PURPOSE: To obtain an assay reagent composed of an alkaline phosphatase labeled with a low-molecular compound and an acridinium salt, having excellent affinity to hapten and high storage stability, reliability and determination sensitivity and useful for chemoluminescent immunoassay, etc. CONSTITUTION: The objective assay reagent having excellent affinity to an antibody having a hapten-polymer composite material as an immunogen and usable e.g. for chemoluminescent immunoassay having improved determination sensitivity is produced e.g. by reacting an acridinium salt [e.g. 10-methyl-N-tosyl- N-(2-carboxyethyl)-9-acridinium carboxamide] with an alkaline phosphatase in the presence of N-hydroxysuccinimide(NHS) and 1-ethyl-3-(3- dimethylaminopropyl)-carbodiimide(EDC) and reacting the obtained alkaline phosphatase conjugate labeled with the acridinium salt with a low-molecular compound (e.g. 3,5,3'-triiodothyronine) in the presence of NHS and EDC. |
