PROBLEM TO BE SOLVED: To reduce an operating condition and a working condition with a less time required for adjusting a column, without a plunger inclining. SOLUTION: A low pressure chromatography column 1 having a diameter of larger than 30cm, is composed of a substrate 12, and a vertical cylinder 4 having a lower sintered disc 3 located above the substrate 12, and an upper sintered disc 16 fixed to the lower side of a lid which is designed as a movable plunger 4. A machine element 5 for vertically moving the plunger 4 incorporates a sensor for determining the vertical posture and the horizontal posture of the plunger 4, and the sensor 6 is electrically connected to a control unit 10 for the machine element 5. A plunger/gel substance can be precisely prepared over the entire range of the plunger by mans of an optical observing unit (endoscope). The column 1 is suitable in particular for refining an insulin and a precursor.
