PURPOSE:To stabilize a protease useful for medicines, cosmetics, detergents and other industrial uses, etc., without impairing the activity of its technical product, by chemically modifying the amino group of a protease with a dicarboxylic acid. CONSTITUTION:A protease derived from an alkali affinitive Bacillus is dissolved in a 0.1M boric acid buffer solution (pH 8) followed by adding a dicarboxylic anhydride (e.g. succinic anhydride) to the solution while keeping the solution at pH 8 with 1N NaOH and a reaction is conducted for 2h to effect chemical modification of the amino group of the protease with the dicarboxylic acid followed by repeated dialysis against a 0.1M phosphoric acid buffer solution at pH 7 to effect the purification of the protease, thus efficiently stabilizing the protease to be used for medicines, cosmetics, detergents and other industrial fields without impairing the activity of its technical product. |
