PURPOSE:To provide a separating medium composed of a polysaccharide derivative supported with silica gel surface-treated with a specified silylating agent. If this separating agent is used in the reverse phase liquid chromatography, this chromatography is excellent in stability of the base line in the mobile phase and shows a high detection peak. CONSTITUTION:This separating agent is prepared by supporting a polysaccharide derivative (preferably, e.g. an ester derivative of cellulose having 10 to 500 number-average degree of polymerization) on silica gel in which the surface silanols are treated with an aralkyl-containing silylating agent {e.g. a compound of formula I [0ne or more of X, Y and Z are C1 or a 1 to 12C alkoxy and the remainders are methyl or ethyl; (n) is 1 to 8; R is a 1 to 4C alkyl or an alkoxy; (m) is 0 or 1; formula II is a 6 to 14C aromatic group]]. The particle size and the average pore size of the carrier silica gel are preferably 1 to 300mum and the 50 to 50000 Angstrom , respectively. In addition, the above-mentioned treatment agent is obtained, e.g. by reacting silica gel with the above-mentioned silylating agent while refluxing a solvent, then mixing the resultant surface-treated silica gel in a polysaciharide derivative solution and subsequently distilling off the solvent.
