PURPOSE:To stably obtain a red natural pigment useful as a colorant for foods, cosmetics, etc., with a high efficiency without being influenced by the natural environment by recovering the red pigment from a cultured cell capable of producing the red pigment of a plant belonging to the genus Aralia. CONSTITUTION:This method for producing a red natural pigment is to take out a leaf and a stem of Aralia cordata Thunb. as a plant belonging to the genus Aralia, wash the taken out leaf and stem with water, sterilize the washed leaf and stem with 70% ethanol and a 10% aqueous solution of bleaching powder, cut the resultant aseptic tissue, bed the cut tissue on an agar culture medium containing an auxin such as 2,4-dichlorophenoxyacetic acid and a cytokinin such as kinetin, culture the tissue in the dark under temperature conditions of 25 deg.C for 2 weeks, provide a callus, then transplant the resultant callus to a culture medium containing coconut milk, culture the callus, induce a cell strain capable of highly producing the red pigment in a part of the callus, then subculture the cell strain, proliferate the cell strain, harvest the cultured cell, freeze-dry the harvested cell, immerse the freeze-dried cell in methyl alcohol acidified with 0.1% hydrochloric acid a whole day and night, extract the pigment, filter the extract solution and concentrate the prepared filtrate under reduced pressure. |
