Detection of an enzyme-specific restriction site in a target DNA sequence comprises: (a) hybridising a nucleic acid fragment contg. the target sequence with an oligonucleotide probe to form a hybrid contg. the restriction site (if present in the target sequence) and a detectable label; (b) immobilising the hybrid on a solid support so that the restriction site (if present) is between the end bound to the support and the portion bearing the label; (c) treating the hybrid with the restriction enzyme in question; and (d) qualitatively or quantitatively determining at least one of the prods. of the enzymatic reaction. Also claimed is a test kit comprising all the reagents required to carry out a process as above.
