<p>Biologically active recombinant apolipoprotein A1 (apoA1) is produced in a T7 RNA polymerase/promoter bacterial expression system that optimizes yield and facilitates subsequent purification of the protein. ApoA1 cDNA is cloned into an Escherichia coli plasmid containing a bacteriophage T7 RNA polymerase promoter; the recombinant plasmid is introduced into an E. coli strain containing inducible bacteriophage T7 RNA polymerase; T7 RNA polymerase is induced in the strain; and the resulting expresses apoA1 is isolated from the culture. The apoA1 protein may be a fusion protein that includes an additional sequence that facilitates purification. Recombinant apoA1 is useful in the treatment and in the prevention of various circulatory related diseases.</p> |
