PURPOSE:To provide a method for producing a saccharide with trehalose, capable of simply and directly producing trehalose from glucose in a high yield and at an extremely low cost without generating by-products, where in the trehalase can safely be used in a food. CONSTITUTION:This novel trehalase hydrolyzes alpha,alpha'-trehalose, 2,2'dideoxy-alphaalpha'- trehalose and 2-deoxy-alpha,alpha'-trehalose into the composing saccharides, respectively but does not act on neotrehalose, lactose, maltose, cellobiose, and sucrose; it has the optimal pH at 5-6, the optimal temperature at 65 deg.C, good thermal stability up to 65 deg.C, a mol.wt. of 400,000-500,000 measured by a gel filtration methoda subunit mol.wt. of 180,000-250,000, the subunit being obtained by an polyacrylamide, an isoelectric point of 2.7 measured by an isoelectric focusing electrophoresis method, and has the characteristics of a sugar protein. |
