<p>PURPOSE:To efficiently prepare a cytoplasmic hybrid callus for breeding, etc., by fusing a protoplast of alfalfa cell obtained by breaking only its cell nucleus in a state leaving useful gene character with a protoplast of the alfalfa cell obtained by inactivating only its cytoplasm. CONSTITUTION:A protoplast obtained by selectively breaking only cell nucleus among protoplasts of alfalfa cell containing a useful gene character in a cytoplasm is used as either one parent and a protoplast obtained by inactivating only cell nucleus among protoplasts of alfalfa cell containing a useful gene character in nucleus is used as the remainder parent and these two kinds of protoplast are fused by applying voltage between parallel electrodes using an electric fusion method, etc., to efficiently provide a hybrid callus and a substituent substance of cytoplasm having useful gene characters in the nucleus and the cytoplasm and capable of utilizing for breeding, etc.</p> |
