The fermentation method of a gene recombination bacillus is characterized by gradually adding a medium contg. a yeast extract and a glucose solution at the high density culture of the bacillus generated from E. coli, and then producing the gene recombination protein. The primary medium is composed of glucose, KH2PO4, MgSO4.7H2O and yeast extract, and the supplying medium is composed of FeSO4.7H2O, CaCl2.7H2O, MnSO4.5H2O, CoCl2.6H2O, ZnSO4.7H2O, CuCl2.5H2O and Na2MoO4.2H2O. The fermentation method inhibits the revelation of the protease, and then prevents the reduction of the revelation ratio per the cell.
