PURPOSE: To enable diagnosis of Alzheimer's disease(AD) by comparing a relative ratio of 76kDa and 80kDa isoforms of calcium-dependent neutral protease (μCANP) in a sample with a control group of a normal individual. CONSTITUTION: A sample such as a frontal lobe cortex sampled from an individual with a doubt of AD is homogenized in a physiological buffer solution, a supernatant liquid obtained by centrifugal separation is poured into a column, andμCANP is eluded by increasing concentration of the buffer solution containing salt. Then,μCANP is given gel-electrophoresis to separate it to 76kDa isoform (activated type) and 80kDa isoform (precursor type), they are made to react on a detectable antibody with immunoreactivity, amounts of antibodies bonded with each of 76kDa and 80kDa are measured to determine relative values, and a relative ratio is obtained by dividing the relative value to 76kDa by the relative value to 80kDa. And when this relative ratio is larger than a control group of a normal individual, the individual with a doubt of AD is confirmed as AD. |
