A method is provided for preselecting a gene or nucleic acid sequence of interest from other DNA, to facilitate further genetic analysis of the preselected DNA. The method comprises an affinity-based process utilizing immobilized affinity molecules to capture hapten-labeled or target molecule-labeled oligonucleotides which have been, or are to be, base-paired by hybridization with the gene or nucleic acid sequence of interest. The method may provide for increased efficiency in the selection of specific recombinant nucleic acid molecules from a larger population of recombinant molecules, and for screening of transformants for those that contain the preselected gene or nucleic acid sequence of interest.
