PURPOSE:To provide a method for simply testing constituent lactic acid bacteria of a lactic acid bacterial starter used in producing a fermented dairy product such as a cheese. CONSTITUTION:This method for testing a starter is to add arginine, Bromocresol Purple(BCP), calcium citrate and carboxymethyl cellulose(CMC) to a culture medium for growing lactic acid bacteria, inoculate the starter thereinto, add 5-bromo-4-chloro-3-indolyl-beta-D-galactopyranoside (Xgal) to the grown colony, judge the presence of Leuconostoc mesenteroides subsp. cremoris by a blue color produced by enzymic reaction, then culture a plate inoculated with lactic acid bacteria at 37 and 25 deg.C, count the constituent lactic acid bacteria from the characteristic of the colony grown in each plate and determine the constituent ratio of the lactic acid bacteria in the starter. |
