PRODUCTION OF COLORED GELATIN AND PI MARKER FOR ISOELECTRIC ELECTROPHORESIS

PURPOSE:To obtain a PI marker enabling easy confirmation of the state of the PI marker in an isoelectric point electrophoresis to enable the accurate determination of collagen and nucleic acid. CONSTITUTION:An amino acid residue of gelatin is chemically modified with a pigment compound at the reactive terminal group other than the color- developing part to effect the coloring of the gelatin and obtain a colored gelatin resistant to discoloration. A PI marker produced by coloring a protein molecule such as decomposed gelatin by similar method enables easy confirmation of the position of the PI marker during or after the electrophoresis. Accurate determination of the isoelectric point of a fibrous protein such as collagen and nucleic acid can be performed by using a PI marker produced by adjusting a decomposed gelatin having an average molecular weight Mw of 15,000-5,000 to a prescribed isoelectric point by chemical modification.
