PURPOSE:To provide new cyclodextrin glucanotransferase obtained from a cultured product of a microorganism which belongs to the genus Bacillus and capable of producing cyclodextrin glucanotransferase, having excellent alkali resistance and useful for detergent, etc. CONSTITUTION:A microbial strain which belongs to the genus Bacillus and capable of producing cyclodextrin glucanotransferase [e.g. Bacillus sp-YT-1 (FERM P-13877)] is cultured and the substance accumulated in the cultured product is separated to obtain the objective enzyme having the following properties. It decomposes starch and synthesizes alpha-, beta- and gamma-cyclodextrin; the optimum pH is about 6 (% soluble starch, 10mM buffer solution, 40 deg.C, 30min); the optimum temperature is about 65 deg.C [1% soluble protein, 10mM phosphate buffer solution (pH10), 10min[; it is stable at pH5-10 (10mM buffer solution, 25 deg.C, 3hr), has a molecular weight of about 43,000 (acrylamide electrophoresis) and is inhibited by Hg<2+>, Ag<+>, Cu<2+>, Ni<2+> and Fe<2+>. |
