Novel compounds I wherein (i) the symbols R are the same or different and each is H or OCH 3 , R 1 is H and R 2 is alkyl, cyanoalkyl, dialkylaminoalkyl, phenylalkyl or amino, or NR 1 R 2 is a 1-piperazinyl group or a 4-alkyl-1-piperazinyl group; (ii) the symbols R, R 1 and R 2 all represent H or (iii) one R is halo and the other is H, halo or OCH 3 or together the symbols R represent -OCH 2 -O-; R 1 is H and R 2 is dialkylaminoalkyl and their acid addition and quaternary ammonium salts are prepared by reacting NH 3 or an amine HNR 1 R 2 with a compound II wherein R 3 is 1-3C alkyl or an acid addition salt thereof. Intermediates isolated are 1-methylthio-6,7- dimethoxy - 3,4 - dihydroisoquinoline (A), prepared by reacting 6,7-dimethoxy-3,4-dihydroisothiocarbostyril with methyl iodide; 6,7- dimethoxy - 3,4 - dihydroisothiocarbostyril (B), which is prepared by cyclization of 2-(3,4- dimethoxyphenyl)-ethyl isothiocyanate (C), which in turn is prepared from 2-(3,4-dimethoxyphenyl) - ethylamine and thiophosgene; 1- methylthio - 6,7 - methylenedioxy - 3,4 - dihydroisoquinoline (D), prepared from methyl iodide and 6,7 - methylenedioxy - 3,4 - dihydroisothiocarbostyril (E), which is prepared by cyclization of 2 - (3,4 - methylenedioxyphenyl) - ethyl isothiocyanate (F) (prepared by reacting 2-(3,4- methylenedioxyphenyl) - ethylamine with thiophosgene); 2 - (4 - chlorophenyl) - ethyl isothiocyanate (G), prepared by reacting 2-(4-chlorophenyl)-ethylamine with thiophosgene; 3,4- dihydro - 7 - chloro isothiocarbostyril (H) prepared by cyclization of (G); and 7-chloro-1- methylthio - 3,4 - dihydroisoquinoline (I), prepared by reacting (H) with methyl iodide. Pharmaceutical compositions comprise a compound I in association with a pharmaceutical carrier or coating, are administered orally, parenterally or rectally and are used as antitussive agents and anti-fibrillation agents. |
