<p>PURPOSE:To obtain a new monoclonal antibody capable of specifically recognizing a cell infiltrated in inflammatory tissue and rapidly recognizing the cell infiltrated in the inflammatory tissue and useful for carrying out diagnosis, etc., of inflammation, e.g. diagnosis of progress and extent of inflammation, detection of treating effect, etc., or judgement, etc., on properties of inflammatory reaction. CONSTITUTION:A bovine gamma-globulin is administered as an antigen to skin of a Hartley type white guinea pig and subcutaneous tissue which caused delay type hypersensitivity reaction is removed and finely cut and cleaned to afford a cell infiltrated in inflammatory tissue. Then, this cell is administered to female Balb/c mouse to immunize the mouse and after 3 days from final immunity, spleen is taken out to separate an antibody-producing cell and the cell is fused with a mouse myeloma cell and the fused cell is cultured in HAT medium to afford a hybridoma. The hybridoma is screened to select desired antibody- producing clone and sub-iected to cloning by a limiting dilution analysis and the resultant monoclonal hybridoma is cultured to provide the objective new monoclonal antibody against a cell infiltrated in inflammatory tissue and useful for diagnosis, etc., of inflammation.</p> |
