BIOLOGICAL METHODS FOR THE PREPARATION OF 7-ACA AND 7-ADAC

The biological methods are effective and economical and are used for producing important intermediate compounds for the preparation of cephalosporin antibiotics, especially 7-aminocephalosporic acid (7-ACA) and 7-amino-deacetylsporic acid (7-ADAC). A transformed strain of Penicillium chrysogenus is cultivated in the presence of adipatic nutrient for the production of adipoyl-6-APA (6-amino-penecillinic acid), followed by in sito expression of the following genesby which P. chrysogenum has been transformed: gene expandase supplying adipoyl-7-ADCA; gene hydroxylase supplying (7-ADAC) and gene acetyltransferase supplying adipoyl-7-ACA. The end product 7-ACA is produced by splitting of the adipoyl side chain by using adipoylacylase.
