PRODUCTION OF GROWTH HORMONE

PURPOSE:To produce a human growth hormone having 20000 daltons molecular weight by efficiently secreting and accumulating the growth hormone proved to deteriorate the diabetogenic activity and leukemogenicity as compared with those of the presently used human growth hormone having 22000 daltons molecular weight in an Escherichia coli periplasm. CONSTITUTION:A human growth hormone secretion plasmid having a DNA fragment containing a promoter taking part in the expression of a neutral protease gene of Bacillus.amyloliquefaciens, a ribosome binding site and a mature human growth hormone gene, capable of coding a human growth hormone having 20000 daltons molecular weight and bound to a site just after a region capable of coding a secretion signal is constructed and then transduced into Escherichia coli. The resultant transformant is subsequently cultured to efficiently secrete and produce a human growth hormone having 20000 daltons molecular weight in an Escherichia coli periplasm.
