This invention generally provides for a method of producing recombinant large latent TGF- beta by introducing nucleic acid sequences coding for LTBP and pro-TGF- beta into eukaryotic cells. This invention further provides for a method for producing recombinant LL-TGF- beta by introducing a DNA sequence for LTBP to an eukaryotic cell which expresses pro-TGF- beta . The invention also provides for the construction of a pME-TGF- beta 2 plasmid and its co-transfection with pRSVneo plasmid into BP-1-1 CHO cells which contains an amplified LTPBP cDNA sequence in the genome. The invention also provides for a method for treating the LL-TGF- beta 1 complex produced to bind to Ca2<+> ions during the purification process activity. An antibody to the large latent TGF- beta 1 complex is also disclosed herein. Additionally isolated L-LAP and a method to produce the same is provided herein.
