PURPOSE:To develop a method for remarkably solubilizing lignin with a bacterium and vigorously fermenting cellulose and proteins, rapidly decompose a hardly decomposable organic substance as its application and effectively utilize the decomposed substance thereof. CONSTITUTION:The strong decomposition and solubilization of a natural fibrous substance or hard proteinaceous material consisting essentially of hardly decomposable lignin or cellulose are facilitated by using a carotenoid-based yellow coloring matter and a strain Thermus aquaticus biovar. SK542 capable of producing a proteolytic enzyme having the high-temperature wide area acting hydrogen ion concentration activity and further utilizing a symbiotic mixed culture of the strain with a new bacterial strain of thermophilic cellulolytic bacterium Clostridium thermocellum biovar.
