<p>PURPOSE:To obtain glycerol derivative, composed of a glycerol derivative containing carboxyl, amino or thiol, etc., capable of introducing a linker or a nonisotopic label into an optional position of DNA, RNA, etc., and useful for detecting, etc., a probe. CONSTITUTION:1,3-Dibenzylglycerol is dissolved in a solvent such as acetonitrile and the resultant solution is then made to react with a compound expressed by the formula X(CH2)n-1Y (X is halogen, Y is halogen or nitrile; (n) is 2-20] in the presence of a base such as butyl-lithium at -80 to +80 deg.C temperature. The resultant reactional product is purified by a silica gel column, etc., and then converted into a carboxylic acid derivative by hydrolysis, etc. A compound expressed by the formula H2N(CH2)mNH2 [(m) is 2-20], HO(CH2)mOH or HS (CH2)qNH2 [(q) is 2-20] is suitably allowed to react with the resultant carboxylic acid derivative to afford the objective derivative expressed by the formula (R<1> and R<2> are H or protecting group of hydroxyl group; R<3> is carboxyl, amino, thiol, etc., which may have a protecting group or a nonisotopic label; L is alkylene containing one or more O, S, NHCO or, CONH bonds).</p> |
