PURPOSE:To produce the prolyl endopeptidase originated from Aspergillus oryzae FS1-32 (FERM 12193). CONSTITUTION:Aspergillus oryzae FS1-32 (FERM 12193) is cultured in a medium containing tannic acid to produce the prolyl endopeptidase. The obtained prolyl endopeptidase has an optimal pH of 5 and an optimal temperature of 37 deg.C, is stable in a pH range of 4-7, and keeps the activity of approximately 80% even when heated at 52 deg.C at a pH of 5 for 1hr. Since the obtained prolyl endopeptidase has an optimal pH in an acidic region, the prolyl endopeptidase can be used in the acidic region enabling to inhibit the multiplication of microorganisms on its industrial utilization such as the removal of the bitter tastes of peptides. |
