PURPOSE:To profitably obtain the subject compound useful as a carcinostatic agent, etc., by determining the whole amino acid sequence of the phenomycin, preparing an expression vector containing its DNA fragment on the basis of the determination result, transforming Escherichia coli with the prepared expression vector and subsequently culturing the transformant. CONSTITUTION:An Actinomyces, Streptomyces fervens var, phenomyceticus is rotation-cultured in a medium at 27 deg.C for 72hr, and the culture solution is filtered. The filtrate is applied to a cation exchange column, and the adsorbed substance is eluted and purified by a high performance liquid chromatography. The amino acid sequence of the collected phenomycin is determined, and on the determined amino acid sequence, a gene coding the phenomycin having the amino acid sequence of the formula is synthesized with a DNA synthesizing machine. The synthesized gene is multiplied by a PCR method, combined with a vector and subsequently introduced into Escherichia coli. The produced transformant is cultured to massively and profitably produce the objective phenomycin of the formula. |
