<p>PURPOSE:To obtain the title protoplast for producing coloring matters for foods or medicines by converting the pistil tissue redifferentiated from saffron callus into protoplast to produce cells of high coloring matter productivity as well as high proliferation rate. CONSTITUTION:The outer skin of the bulbs of saffron with its sprouts grown to ca. 10cm is peeled off and discolored parts are removed, followed by washing with running water for a few hours and then successive treatment with 10% benzalkonium chloride, 70% ethanol, and 5% sodium hypochlorite, in this order. Then, buds are taken out from the sprouts and cleaved, and the pistil is taken out and inoculated into a medium followed by addition of naphthaleneacetic acid and benzyladenine, and the resulting pistil redifferentiated from the callus is collected. This pistil is then shaken at 30 deg.C for one hour in an enzyme solution containing e.g. cellulase followed by centrifugation of such portions as to be passable through a 200-mesh nylon net, thus obtaining the objective protoplast.</p> |
