<p>The presence of soluble endothelial leukocyte adhesion molecule (ELAM) in body fluids is an indicator of the state of activation of, and/or damage to, the endothelium. Homogeneous preparations of soluble ELAM, which are useful as assay standards in diagnostic and prognostic assays for diseases affecting the vasculature (particularly complications of diabetes leading to retinopathy), are prepared in a variety of ways, including detergent extraction from cells expressing membrane-bound ELAM and expression of recombinant DNA coding for a truncated form of ELAM. The truncated, soluble ELAM comprises the lectin domain and at least part of (and preferably all) the EGF-like domain. Affinity chromatography forms a useful final purification step.</p> |
