<p>PURPOSE:To obtain the subject new derivative, having rings A and C and useful as a raw material for compounds having vitamin D activity by using an olefin compound as a starting raw material, providing a conjugated olefin compound which is an intermediate and further reacting the resultant compound. CONSTITUTION:An olefin compound expressed by formula I (R<1> and R<2> are H or protecting group of OH) is subjected to halogenating reaction to provide a compound expressed by formula II (X is halogen), which is then reacted with lithium diphenylphosphide. The resultant compound is then treated with an acid to afford a compound expressed by formula III (Ph is phenyl), which is further reacted with cyclohexanone to provide a conjugated olefin compound expressed by formula IV. The prepared compound expressed by formula IV is then oxidized to afford a compound expressed by formula V, which is subsequently reacted with nitromethane to provide a compound expressed by formula VI. The obtained compound expressed by formula VI is further cyclized to afford a compound expressed by formula VII, which is then reductively hydrolyzed to provide a compound expressed by formula VIII, e.g. 6-(t-butyldimethylsilyloxy)-4-(t-butyldiphenylsilyloxy)-2-[(2-cyclohex ylidene-1- hydroxy)ethyl]cyclohexanone.</p> |
