RECOMBINED ACARIDAL ALLERGEN

PURPOSE:To provide a safe and active recombined acaridal allergen having an epitope contained in an acaridal allergen originated from acarids, free from impurities inducing anaphylaxis, and useful as an agent for treating and diagnosing acaridal allergic diseases. CONSTITUTION:The objective recombined acaridal allergen containing an amino acid sequence of the formula is obtained by the steps of grinding living acarids prepared by culturing and multiplying Dermatophagoides farinae in a 5.5M isocyanic acid guanidine solution; subjecting the ground product to centrifugal separation; collecting the supernatant; extracting all RNA fractions from the supernatant by a conventional method; applying the fractions to an oligo (dT) cellulose column to isolate mRNA, using the mRNA as a template to synthesize a CDNA library; cloning the library to screen a DNA coding the acaridal allergen; treating the DNA with a restriction enzyme; bonding the product to a vector, introducing the vector into a host such as bacteria, yeast or mammalian cells; and subsequently culturing the transformant.
