PURPOSE:To purify enzyme on an industrial scale by bringing an extracted solution of a specific enzyme into contact with an adsorbent having protein bonded dye linked to a supporting carrier, adsorbing only the enzyme to be specifically bonded to the dye and then eluting the adsorbed enzyme. CONSTITUTION:Cells of bacterium of Zymomonas mobilis are crushed by mechanical grinding, the prepared extracted solution of enzyme is brought into contact with an adsorbent of affinity chromatography having protein bonded dye such as chlorotriazine group-containing reactive dye, only the enzyme such as alcohol dehydrogenase to be specifically linked to the protein bonded dye is selectively adsorbed and then the adsorbed enzyme is eluted to purify the enzyme. |
