PURPOSE:To obtain inexpensive L-proline in high yield at a high accumulation concentration by culturing a specific microorganism, accumulating the L-proline in a culture solution and collecting the L-proline from the above-mentioned culture solution. CONSTITUTION:A microorganism [Providencia.rettgeri PX8.5-13 (FERM P-11936)], belonging to the genus Providencia and having the tolerance to aspartic acid metabolic antagonistic substances and the ability to produce L- proline is cultured in a liquid culture medium at pH5-7 and 24-37 deg.C temperature for 4-120hr by shaking culture, etc., and the resultant culture solution is then filtered to regulate the filtrate to pH2. The regulated filtrate is subsequently passed through and adsorbed on a weakly acidic cation exchange resin. Adsorbed ingredients are subsequently eluted with a dilute aqueous ammonia and deammoniation is carried out to concentrate the deammoniated ingredients and afford the objective L-proline. |
