PURPOSE:To enable simple obtaining of the subject enzyme by culturing a specific strain, extracting and purifying the resultant microbial cells. CONSTITUTION:A strain such as Alcaligenes.faecalis capable of producing diacetylpolyamine amidohydrolase (APAH) is cultured in a culture medium containing glucose, etc., at 15-40 deg.C and pH 5.0-9.0 by spinning culture with aeration to provide a culture. Microbial cells in the resultant culture are then extracted and subsequently purified by a gel filtration method, etc., to produce the objective diacetylpolyamine amidohydrolase having the following physico- chemical properties. Action; hydrolyzing amide bonds of diacetylpolyamines and producing a monoacetylpolyamine. Substrate specificity; acting on the diacetylpolyamines such as diacetyl-1,6-diaminohexane, diacetylptrescine or diacetylcadaverine. Optimum pH; pH 8.5-9.5. pH stability; having >=90% residual activity at pH 7.0-9.0 in preserving at 30 deg.C for 30min, etc. |
