PURPOSE:To enable simple obtaining of the subject enzyme by culturing a specific strain, extracting and purifying the resultant microbial cells. CONSTITUTION:A strain such as Candida.guilliermondii having the ability to produce diacetylpolyamine amidohydrolase is cultured in a culture medium containing molasses, etc., at 15-40 deg.C and pH 5.0-9.0 by stirring culture with aeration to provide a culture, which is then centrifuged to afford microbial cells. The obtained microbial cells are extracted and subsequently purified to produce the objective diacetylpolyamine amidohydrolase having the following physico-chemical properties. Function; hydrolyzing amide bonds of diacetylpolyamines and producing a monoacetylpolyamine. Substrate specificity; acting on the diacetylpolyamines such as diacetyl-1,2-diaminoethane, diacetylptrescine or diacetylcadaverine. Optimum pH; pH 7.0-8.0. pH stability; having >=90% residual activity at pH 5.5-11.3 in preserving at 30 deg.C for 30min, etc. |
