PURPOSE:To efficiently improve environmental hygiene by searching for and separating a microorganism capable of decomposing fish scale collagen from soil. CONSTITUTION:A strain separated from soil is purified and subsequently cultured in a liquid culture medium containing sterilized fish scales to afford a culture. A strain having fish collagen decomposing activity is then screened from the resultant culture to afford Micrococcus roseus SC-002 strain having the following aerobic bacteriological properties. Morphological findings; Gram- positive coccus of 1.0-1.5mum size exhibiting a single cell or linkage form by culturing at 30 deg.C for 48hr without any motility and sporulation. Culture findings; red, circular, smooth and opaque colonies by culturing at 30 deg.C for 48hr in a bouillon agar plate culture medium without growing in a glucose agar culture medium. Physiological findings; reducing ability of nitrates; positive, urease; negative and starch hydrolyzing ability; positive. Growth range; pH6-10. Ortimum temperature; 30 deg.C. Utilizing glucose without utilizing galactose. Chemical properties; 67.4mol% content of (guanine + cytosine), etc. |
