NEW MATERIAL:Bone morphogenetic protein(BMP) of Xenopus laevis. USE:A therapeutic agent of bone fracture and osteoporosis. PREPARATION:For example, liver of Xenopus laevis is powdered in a liquid nitrogen to afford liver powder and a buffer containing a proteinase is added to the liver powder, the powder is incubated at 50 deg.C for 2hr. DNA sample therefrom is treated with phenol, then phenol is removed by dialysis and RNA is removed by carrying out RNAse treatment, and the treated DNA sample is dialyzed to afford chromosomal DNA derived from the liver. Then the chromosomal DNA is partially cut with a restriction enzyme and the cut DNA is bound to a phage DNA which becomes a vector to prepare a DNA library. Escherichia coli is in fected with the vector and subjected to screening using a probe to select out a positive clone and DNA is extracted therefrom. The DNA is then subjected to restriction enzyme treatment to bind the DNA to the vector and the vector-bound DNA is inserted into Escherichia coli to transform the bacilli and the transformant is cultured to provide the MBPs protein. |
