<p>NEW MATERIAL:A DNA sequence containing a base sequence of the formula, also containing the promoter and/or protein-coding region of glucoamylase gene isolated from Aspergillus oryzae. USE:Production of glucoamylase, saccharifying the starch to be used for wine brewing and saccharide production. PREPARATION:For example, mRNA is extracted, by a conventional means from the microbes of Aspergillus oryzae incubated in a medium containing starch as the sole carbon source, and using the mRNA as template, cDNA is synthesized to produce cDNA library, and then, using a probe, a clone containing DNA coding glucoamylase is selected by plaque hybridization technique. From this clone, DNA is extracted and put to restriction enzyme treatment, and then subjected to agarose gel electrophoresis to carry out purification. The resultant DNA is bound to a vector, thus obtaining the objective gene of the formula.</p> |
