PURPOSE:To enable obtaining of an extremely powerful hybrid promoter sequence by using an upstream control sequence, derived from Escherichia coli and capable of functioning in a yeast. CONSTITUTION:An upstream control sequence(URS) is obtained from an Escherichia coli chromosomic DNA. In order to excise such a sequence from the Escherichia coli chromosomic DNA, a control sequence is obtained by cleaving of nonspecific double strands with DNaseI or complete hydrolysis of a DNA with Sau3AI in which the recognition sequence is 4 base pairs. In addition to the aforementioned ADHI promoter, GAPDH promoter, PGK promoter, PYK promoter, ENO1 promoter, etc., are cited as the yeast promoter capable of functioning in combination with the upstream control sequence. GAPDH promoter, PGK promoter, ADHI promoter, etc., are cited as a TATA region of the hybrid promoter composed of the upstream control sequence and the yeast promoter. |
