<p>PURPOSE:To enable the mass production of lemon balm through producing its protoplast by isolating it in an enzyme solution containing specific pharmaceutical preparations etc., using the callus obtained by incubating lemon balm seeds in an auxin-contg. medium. CONSTITUTION:Firstly, lemon balm seeds are sterilized and incubated in an e.g. auxin-contg. MS medium in a sterilized state to obtain callus. Thence, the objective protoplast can be obtained in several hours by isolation from the above callus in an enzyme solution containing cellulase pharmaceutical preparation, pectin lyase, polygalacturonase pharmaceutical preparation, CaCl2.2 H2O, and, an osmotic pressure regulator, succharide such as sorbitol, mannitol or sucrose. In this case, the respective optimal levels of the components in this enzyme solution are set for the reason of activity maintenance. Thereby, the present protoplast can be isolated in a short time, and because of rapid growth of the callus, the above protoplast can be always grown artificially in all seasons in high reproducibility, leading to the possibility of the mass production of new kinds of lemon balm.</p> |
