MONOCLONAL ANTIBODY AGAINST REVERSE TRANSCRIPTASE OF HUMAN IMMUNODEFICIENCY VIRUS, ITS PRODUCTION AND STRAIN CAPABLE OF PRODUCING THE SAME ANTIBODY

PURPOSE:To produce a monoclonal antibody capable of inhibiting the reverse transcriptase activity of a human immunodeficiency virus by culturing a hybridoma strain prepared by specific cell fusion. CONSTITUTION:A mouse is immunized with a vaccinia virus containing a gene capable of coding a reverse transcriptase (RT) region of a human immunodeficiency virus (hereinafter referred to as HIV) transduced thereinto and lymphocytes obtained from the spleen thereof are fused to mouse myelomatous cells to afford a hybridoma strain. The resultant strain is then cultured, selected and cultured in a flask or in an animal abdominal cavity. A monoclonal antibody having a high antibody value inhibiting the RT activity of the HIV is contained in the culture supernatant or ascitic supernatant obtained by centrifugal operation. The prepared antibody is subsequently purified by applying a normal method to afford a high-purity product. Thereby, the resultant monoclonal antibody can be expected as a reagent for fundamental research on HIV, a test reagent and a remedy for AIDS.
