NEW MATERIAL:DNA coding an alpha-1,3-glucan 3-glucanohydrolase enzyme protein produced by Trichoderma harzianum. USE:Mass-production of alpha-1,3-glucanase, etc. PREPARATION:The amino acid composition and the N-terminal amino acid sequence of alpha-1,3-glucanase are analyzed and an anti-alpha-1,3-glucanase serum is prepared. A cDNA library is prepared from the microbial cell, alpha-1,3-glucanase cDNA is screened, the longest cDNA among the cloned cDNA is subjected to sub-cloning and a transformant cloned with the cDNA fragment is separated. The objective alpha-1,3-glucanase cDNA is produced from a plasmid DNA separated from the transformant. |
