A method for isolation and purification of human growth hormone (hGH) in yeast cells comprises (a) suspending and crushing the hGH expressed yeast cells in buffer soln., increasing pH, and removing the residue, (b) decreasing pH to remove the soluble proteins, increasing pH to dissolve proteins, and then decreasing pH to 5.0- 6.5 to remove the precipitate and obtain supernatant, (c) purifying it by anion exchange chromatography, gel chromatography, anion exchange chromatography and hydrophobic gel chromatography, and (d) solvent-dialyzing it in glycine/sodium phosphate mixt. soln.
