NEW MATERIAL:A DNA containing a gene region coding cytidine 5'- triphosphate-synthesizing enzyme derived from the chromosome DNA of Bacillus bacteria giving cytidine analogue resistance. USE:Preparation of cytidine useful for e.g. raw materials for synthesizing biochemical reagents and medicines. PREPARATION:For example, Bacillus subtilis DFCR-100 strain giving cytidine analogue resistance and capable of producing cytidine is inoculated into a culture medium and incubated, and the resulting microbes are collected and put to DNA extraction by a conventional means, thus obtaining a chromosome DNA. Thence, this DNA is put to restriction enzyme treatment, bound to a vector and inserted into Escherichia coli, and the resulting transformant is incubated to select clone with cytidine 5'-triphosphate-synthesizing activity. From this clone, plasmid is taken out and put to restriction enzyme treatment, thus obtaining the objective DNA containing a gene region coding cytidine 5'-triphosphate-synthesizing enzyme. |
