Chondroitin- sulphate is prepd. from 1-1.8 pts. wt. of bovine tracheae. These are digested in two stages with 0.5-0.55 M sodium hydroxide for 24 hrs. at 4-5 deg.C., followed by filtration and addn. of 0.20-0.30 pts.wt. of celite. Following a 24 hrs. rest in the cold, the mixt. is filtered, pH is set to 7-7.25 and 1.25 vols of ethanol are added, and the mixt. is allowed to stand in the cold for 24 hrs. The mixt. is centrifuged. The ppte. is dissolved in distilled water and after 24 hrs standing in the cold, further 2 ools of ethanol, contg. 0.5% sodium acetate, are added. The ppte. obtd. by centrifuging is dried with ethanol or di ethyl-ether until formation of a powder is observed.
