Determination of protease inhibitor (I) comprises mixing a (I)-contg. liq. with a protease substrate (I), then starting the reaction by adding the appropriate protease (III). The rate of hydrolysis of (II) is then measured. Pref. the substrate is a peptide of formula A-B-C-D-R (A=residue of free or protected amino acid; conventional N-terminal protecting gp. or H; B,C and D = residue of free or protected amino acid; R = chromogenic, fluorogenic or luminogenic residue which can be determined by photometry, fluorimetry or luminescence measurement, or it is the residue of a mercaptan which after hydrolysis of the D-R bond can be photometrically determined by conversion to a dye).
