<p>PURPOSE:To obtain a reagent for examination and diagnosis of cell, body fluid, etc., by forming a complex with an antigen derived from Cryptococcus uniguttulatus. CONSTITUTION:A bacterium cell obtained by culturing Cryptococcus uniguttulatus JCM3685 strain is in a culture medium is subjected to lytic reaction to afford an antigen liquid (A). Then spleen is delivered from a mouse immunized with the liquid A to provide an immunized spleen cell (B). Then mouse myeloma cell (C) such as P#U1 is blended with the cell B at a ratio of cell number of the ingredient C: the ingredient B of 1:1-10 and the blend is fused with electric fusion method, etc., to provide the hybridoma (D). Then the ingredient D is subjected to screening with enzyme immune measuring method, etc., to afford a fused cell (e.g. 1CrU39-52) (E). Then the cell E is cultured with HAT culture medium containing glutamine, etc., to produce the aimed monoclonal antibody.</p> |
