NEW MATERIAL:A DNA fragment containing a gene to order etherase among a fragment obtained by cleaving chromosome DNA, of a bacterium to produce an enzyme (etherase) to specifically cleave arylglycerol-beta-aryl ether type bond, a main intramolecular bond of lignin, with a restriction enzyme. USE:Producing a large amount of lignin hydrolyzing enzyme. PREPARATION:For example, chromosome of DNA of wild strain (GYK-6 strain) of bacterium Pseudomonas paucimobilis capable of producing etherase is extracted by a conventional procedure, cut out with restriction enzyme SalI to form fragments, from which DNA fragment containing five SalI cleaving sites, coding etherase gene to be divided into 7.0, 4.4, 3.0, 2.6, 1.9 and 1.6Kbp fragments and having 20.5Kbp whole length. |
