Test for distinguishing between pigs that are infected with Aujeszky's disease virus and pigs that have been vaccinated against the disease, where the infective form of the virus contains an antigenic determinant (AD-1) not present in the form used for vaccination, comprises: (a) either (i) incubating a serum sample with the infective form of the virus and introducing the mixt. into an ELISA plate coated with a monoclonal antibody (MAB) specific to one epitope of AD-1, or (ii) introducing a serum sample into an ELISA plated coated with the virus or with a synthetic or non-synthetic antigen or epitope corresp. to the infective or vaccinating form of the virus; (b) incubating the mixt. in the presence of (1) a conjugate of an enzyme A and a MAB specific to another epitope of AD-1 and (2) a conjugate of an enzyme B and a MAB specific to an antigenic determinant (AD-2) present in the vaccinating form of the virus; and (c) washing the plate, incubating with a soln. contg. substrates for enzymes A and B, opt. adding a soln. to stop the conversion of the substrate, and photometrically determining the converted substrates.
|
